The present study evaluated the anti-obesity potential of Trim-fit in a high-fat diet (HFD)-induced obesity model using key parameters including feed consumption, body weight, meal pattern behavior, lipolysis, and oxidative stress markers. HFD-induced obesity is known to cause hyperphagia, excessive weight gain, impaired satiety signaling, reduced fat metabolism, and increased oxidative stress, closely mimicking human obesity and associated metabolic dysfunctions. In this model, Trim-fit treatment demonstrated beneficial effects by modulating feeding behavior, reducing body weight gain, improving meal patterns, enhancing lipolysis, and attenuating lipid peroxidation, thereby indicating its potential role in the management of obesity and related metabolic disturbances. Body Weight : % Change in Body Weight: In the vehicle-treated control group, the percentage increase in body weight was 15.12 ± 2.56%, reflecting normal physiological growth. In contrast, the obese group showed a marked increase to 48.12 ± 2.85%, indicating excessive weight gain and successful induction of obesity. Treatment with metformin significantly reduced the percentage weight gain to 23.46 ± 7.14%, demonstrating an approximate 51% reduction compared to the obese group. Similarly, Trim-fit single dose (T1) treatment further reduced weight gain to 18.49 ± 1.51%, reflecting an approximate 61–62% reduction. Trim-fit double dose (T2) showed a comparable effect, reducing weight gain to 17.44 ± 2.12%, corresponding to an approximate 63–64% reduction. These findings indicate that both Trim-fit treatments, particularly T2, were more effective than metformin in controlling obesity-induced weight gain. Meal Duration Analysis: In the control group, the average meal duration was 3.5 ± 0.4 min/meal, indicating normal feeding behavior. In contrast, the HFD-induced obese group showed a marked increase to 5.8 ± 0.6 min/meal, reflecting prolonged feeding and altered satiety signaling. Treatment with metformin reduced meal duration to 4.7 ± 0.5 min/meal, demonstrating an approximate 18–20% reduction compared to the obese group. Similarly, Trim-fit (T1) treatment decreased meal duration to 4.8 ± 0.5 min/meal, indicating a comparable reduction (~17%). However, Trim-fit (T2) showed a lesser effect, with meal duration of 5.2 ± 0.5 min/meal, corresponding to an approximate 10% reduction. These findings suggest that while all treatments improved feeding behavior, metformin and Trim-fit (T1) were more effective than Trim-fit (T2) in normalizing meal duration. Meal Pattern Analysis: These findings suggest that Trim-fit, particularly at the T2 dose, effectively restores altered meal patterns and improves satiety regulation in obese group. In the control group, meal frequency was 12 ± 1 meals/day with an inter-meal interval of 60 ± 5 min, reflecting normal feeding rhythm and satiety regulation. In contrast, the HFD-induced obese group showed a reduced meal frequency of 8 ± 1 meals/day along with a shortened inter-meal interval of 35 ± 4 min, indicating disrupted feeding patterns and impaired satiety control. Treatment with metformin improved meal frequency to 10 ± 1 meals/day and increased the inter-meal interval to 50 ± 5 min, demonstrating partial normalization of feeding behavior. Similarly, Trim-fit (T1) treatment resulted in 9 ± 1 meals/day with an inter-meal interval of 48 ± 4 min, indicating moderate improvement. Notably, Trim-fit (T2) showed the most pronounced effect, increasing meal frequency to 11 ± 1 meals/day and extending the inter-meal interval to 52 ± 5 min, approaching normal control values. Lipolysis: Notably, Trim-fit (T2) showed the most pronounced effect, increasing lipolysis to 4.7 ± 0.3 nmol glycerol/ml, corresponding to an approximate 51–52% improvement and approaching normal control levels. In the control group, lipolysis levels were 5.2 ± 0.4 nmol glycerol/ml, indicating normal fat breakdown. In contrast, the obese group showed a marked reduction to 3.1 ± 0.3 nmol glycerol/ml, reflecting impaired lipolytic activity associated with obesity. Treatment with metformin increased lipolysis to 4.0 ± 0.3 nmol glycerol/ml, demonstrating an approximate 29% improvement compared to the obese group. Similarly, Trim-fit (T1) treatment elevated lipolysis to 3.8 ± 0.4 nmol glycerol/ml, indicating a moderate increase (~22–23%). These findings suggest that Trim-fit, particularly at the T2 dose, effectively enhances fat breakdown and improves lipid metabolism in obese group. Lipid Peroxidation (MDA Assay): Notably, Trim-fit (T2) showed the most pronounced effect, reducing MDA levels to 2.2 ± 0.2 nmol/ml, corresponding to an approximate 51% reduction and approaching normal control levels. These findings suggest that Trim-fit, particularly at the T2 dose, effectively attenuates oxidative stress and lipid peroxidation in obese group. In the control group, MDA levels were 1.8 ± 0.2 nmol/ml, indicating minimal lipid peroxidation and normal oxidative status. In contrast, the obese group showed a marked increase to 4.5 ± 0.4 nmol/ml, reflecting enhanced oxidative stress and lipid peroxidation associated with obesity. Treatment with metformin reduced MDA levels to 3.2 ± 0.3 nmol/ml, demonstrating an approximate 29% reduction compared to the obese group. Similarly, Trim-fit (T1) treatment decreased MDA levels to 3.4 ± 0.3 nmol/ml, indicating a moderate reduction (~24–25%).
